Experimental study on production of Freeze Dried B.C.G. Vaccine in Korea

Abstract
Author Prepared 30 lots of Freeze dried B.C.G. Vaccine with using Stokes Freeze dryer. The vaccine was dried for 24 hours. in a vacuum of 200 micron at 25 ℃, after few hours initiate freezing at -40 ℃, The number of viable count was determined by the quantitative culture on Löwenstein. Jensen medium. The survival rate was 37% compared before freeze drying and the viavle rate after 3 months. storage at 5℃ was 81 % and 56% after 6 months. ln regard to the tuberculin conversion rate to schoolchildren, the freeze dried B.C.G. vaccine showed 66% comparing with the wet B.C.G. vaccine which showed 76%. And the size of average induration by the freeze dried vaccine was a little smaller than that by the' wet vaccine, the size of the vaccination lesion by the freeze dried B.C.G. vaccine was smaller then that of wet B.C.G. vaccine.
